چکیده

Background Signal transducer and activator of transcription 3 (STAT3) is a factor that belongs to class targets devoid catalytic function, thus deemed “undruggable” by standard modalities such as small molecule inhibitors or biologics. STAT3 can be activated various receptor- non-receptor tyrosine kinases, playing critical role in activation pathways triggered cytokines, hormones, growth factors, making it an attractive target for the treatment inflammatory diseases. Objectives Kymera has developed heterobifunctional molecules selectively degradation elimination ubiquitin-proteasome pathway. We sought evaluate pharmacologic potential these degraders through vitro vivo studies relevant human autoimmune disease, including murine collagen-induced arthritis. Methods evaluated impact on monocytes, dermal fibroblasts, CD4+ T cells, PBMC LPS, IL-6/IL-6R, IL-21, IL-23, well anti-CD3/CD28 plus cocktail cytokines antibodies. pSTAT3 inhibition were determined comparison JAK1/2 inhibitor. Inhibition chemokines, collagen release, Th17 (CD4+CD25-RORγt+CXCR6+) Treg (CD4+CD25+CD127lowFOXP3+) expansion used efficacy assays. Finally, tested vivo, mechanistic (IL-6 challenge) disease model (murine CIA) rheumatology indications. Results showed broad potent activity in-vitro against TLR receptor cytokine-induced immune stromal soluble mediator release MCP-1/CCL2 Collagen1a1. cells robustly inhibited development abrogating IL-17, IL-22, IL-8/CXCL8, TNFα production, increased numbers manner superior inhibition. In mice injected with IL-6, plasma levels serum amyloid A dose-dependently suppressed degradation. arthritis model, resulted robust, dose-dependent delay onset decreased incidence, clinical scores, local cytokine expression (paws) histopathological complete alleviation periosteal bone growth. Conclusion These data demonstrate alleviating inflammation models disease. IL-4 Can Inhibit IL-17 Production in Collagen Induced Arthritis

Background: IL-4 is a cytokine that induces differentiation of naive helper T cells into Th2 cells. Once activated by IL-4, Th2 cells subsequently produce additional IL-4. Objective: To examine the effect of IL-4 on IL-17 production and its effect in Collagen- Induced Arthritis (CIA) mice. Method: In this study, a chicken collagen-II-induced experimental arthritis (CIA) model was used in DBA/1 ...

An herbal formula attenuates collagen-induced arthritis via inhibition of JAK2-STAT3 signaling and regulation of Th17 cells in mice

Wenjinghuoluo prescription, a traditional Chinese medicine compound treatment of rheumatoid arthritis characterized by wind-cold-dampness arthralgia, contains five herbs, namely, C. cassia Presl., Cinnamomum cassia Presl., Paeonia lactiflora Pall., Saposhnikovia divaricate (Turcz.) Schischk., and Clematis chinensis Osbeck. We have reported that WJHL could inhibit the production of inflammatory ...

EGCG Attenuates Autoimmune Arthritis by Inhibition of STAT3 and HIF-1α with Th17/Treg Control

Epigallocatechin-3-gallate (EGCG) is a green tea polyphenol exerting potent anti-oxidant and anti-inflammatory effects by inhibiting signaling and gene expression. The objective of the study was to evaluate the effect of EGCG on interleukin (IL)-1 receptor antagonist knockout (IL-1RaKO) autoimmune arthritis models. IL-1RaKO arthritis models were injected intraperitoneally with EGCG three times ...

Inhibition of microRNA-34a ameliorates murine collagen-induced arthritis

Rheumatoid arthritis (RA) is one of the most frequently occurring autoimmne diseases, with symptoms including synovium hyperplasia, immune disorder, cartilage damage and bone resorption. It has previously been demonstrated that microRNA-34a (miR-34a) may participate in cell apoptosis, immune activation and bone metabolism, therefore the present study investigated the effects of miR-34a on RA. C...
